بیماری تب برفکی یا بیماری دهان و پا یك بیماری عفونی و گاهی کشنده ویروسی است که از جنس آفتوویروس، خانواده پیکورناویریده و در دسته RNA ویروس ها می باشد. این ویروس شامل هفت سروتیپمتمایز از یکدیگر می باشد و عبارتنداز: 3SAT , 2SAT , 1SAT , C , .1Asia , O , A که تمام این سروتایپ های تب برفکی می توانند بیماری را به شکلی ایجاد نمایند که از نظر بالینی غیر قابل تفکیك ولی از نظر ایمنی شناختی با یکدیگر متفاوت باشند. عفونت با هر یك از سروتیپ ها ایمنی در برابر سروتیپ دیگر ایجاد نمی کند. با توجه به اعالم سازمان دامپزشکی کشور، برابر گزارش سرویس دامپزشکی کشور عراق به سازمانجهانی بهداشت دام )WOAH)، سال 2022 نمودار بروز کانونهای بیماری تب برفکی با سویه 2SAT در تمامی استانهای آن کشور روند افزایشی و همراه با تلفات بوده است. همچنین گزارشهای رسمی از افزایش روند بروز بیماری با سویه مذکور درکشورهای ترکیه ،بحرین و عمان، نشان دهنده افزایش خطر نفوذ این سویهاگزوتیك به ایران بود. متعاقب گزارش مبنی بر شیوع بیماری تب برفکی در استان قم بعد از دریافت نمونه های بافت اپیتلیوم زبان گوساله های مشکوك به این بیماری، از بافت ها سالیه تهیه و روی سلول های حساس شامل سلول پرایمری تیروئید گاو (BTY (و دو سلول الین -2RS-IB و BHK تلقیح شد. به دلیل حساسیت بیشتر سلول BTY به ویروس در مرحله اول، سروتیپ 2SAT در پاساژ دوم این سلول بدست آمد و در مراحل بعد سازگاری ویروس به دو سلول الین ذکر شده انجام شد. سپس دو کلون از ویروس 2SAT تهیه و آزمایشات تعیین ویژگی شامل تعیین هویت به روش های االیزا و مولکولی و تعیین توالی و نوع توپوتایپ روی جدایهویروس انجام شد و در ادامه آزمایشات کنترل کیفی شامل استریلیتی، مایکوپالسما، ، خلوص ویروس از نظر عدم آلودگی به سایر سروتیپ های ویروس تب برفکی و عوامل ویروسی ناخواسته مانند ,BTV3,PI ,BVDBLV, Herpes Pan روی جدایه ویروس انجام گردید. دو کلون از ویروس جدا شده به بخش بیوبانك و بخش تولید واکسن تب برفکی در موسسه رازی تحویل داده شد. جدایه ویروس بدست آمده با مشخصات /2024MK-Razi/IRN نام گذاری گردید.کلمات کلیدی: ویروس تب برفکی- سروتیپ – 2SAT -تعیین ویژگی

Foot-and-mouth disease (FMD) or hoof-and-mouth disease is a highly contagious and sometimes fatal viral disease that belongs to the Aphthovirus genus, Picornaviridae family, and is classified as an RNA virus. This virus includes seven distinct serotypes: A, O, Asia.1, C, SAT1, SAT2, and SAT3. All of these FMD serotypes can cause the disease in a clinically indistinguishable manner, but they differ immunologically.Infection with one serotype does not confer immunity against the others. According to the announcement of the Veterinary Organization of the country, based on the report of the Iraqi Veterinary Service to the World Organization for Animal Health (WOAH), in 2022, the chart of the outbreak of FMD with the SAT2 strain showed an increasing trend in all provinces of that country, accompanied by mortality. Official reports of the increasing trend of the disease with the mentioned strain in Turkey, Bahrain, and Oman also indicated an increased risk of the penetration of this exotic strain into Iran.Following the report of the outbreak of foot-and-mouth disease in Qom province after receiving tongue epithelial tissue samples from calves suspected of this disease, tissue slices were prepared from the tissues and inoculated onto sensitive cell lines, including primary bovine thyroid (BTY) cells and two cell lines, IB-RS-2 and BHK.Due to the higher sensitivity of BTY cells to the virus in the first stage, the SAT2 serotype was obtained in the second passage of this cell, and in the subsequent stages, the virus adaptation to the two mentioned cell lines was performed. Then, two clones of SAT2 virus were prepared and characteristic tests including identification by ELISA and molecular methods and determination of sequence and type of topotype were performed on the virus isolate, and then quality control tests including sterility, mycoplasma, purity of the virus in terms of non-contamination with Other foot and mouth virus serotypes and unwanted viral agents such as BVD, PI3, BTV, Pan Herpes, BLV were performed on the virus isolate.Two clones of the isolated virus were delivered to the Biobank and Foot-and-Mouth Disease Vaccine Production Department at the Razi Institute. The obtained virus isolate was named IRN/Razi-MK/2024.Keywords: Foot-and-mouth disease virus - Serotype - SAT2 - Characterization.
